Long intergenic non-protein coding RNA 342 (LINC00342)

Target
LINC00342
Molecular classification
Long non-coding RNA (lncRNA), Other (not protein, not a receptor, not an enzyme; intergenic transcript)
01

Overview

Long intergenic non-protein coding RNA 342 (LINC00342) is a long non-coding RNA transcript located on chromosome 2q11.1, not overlapping any protein-coding regions. It is upregulated in various cancers, including colorectal cancer and non-small cell lung cancer, and acts as an oncogene by promoting cancer cell proliferation, migration, invasion, and tumor growth. Mechanistically, it functions as a competitive endogenous RNA (ceRNA), binding miR-19a-3p and regulating the expression of aminopeptidase like 1 (NPEPL1). High expression of LINC00342 correlates with poor prognosis in several cancers. LINC00342 is emerging as a potential diagnostic and prognostic biomarker, though direct drug targeting has not yet reached clinical use.

Mechanism of action

Antisense oligonucleotide-mediated knockdown (inhibits LINC00342 activity by degrading or blocking its RNA transcript). Small molecule inhibitors or RNAi (theoretical approaches to interrupt its function or expression).

03

Biological functions

Cell proliferation (promotes tumor cell growth in vitro and in vivo)Cell migration (facilitates metastatic phenotypes)Cell invasionGene expression regulation (participates in competitive endogenous RNA networks by sponging miRNAs, notably miR-19a-3p)Other (epigenetic regulation, ceRNA function)
04

Disease associations

Cancer (especially colorectal cancer and non-small cell lung cancer; acts as oncogene)Other: tumor progression, poor prognosis association in NSCLC and infantile hemangiomas
05

Safety considerations

Off-target effects (by virtue of being a non-coding RNA, targeting may affect other cellular RNAs)Unknown physiological roles outside cancer (possible risk of disrupting normal gene regulation if targeted in non-cancer cells)Delivery challenges (typical for lncRNA-targeted therapeutics; issues include cell- or tissue-specific targeting and stability)
06

Biomarkers

Upregulated LINC00342 expression in tumor tissue (potential diagnostic/prognostic biomarker in colorectal cancer and NSCLC)Correlated with poor clinical outcome
