Overview

PRECSIT, also known as LINC00346 and several other aliases, is a long intergenic noncoding RNA (lncRNA) that is upregulated in multiple cancers, most notably cutaneous squamous cell carcinoma (cSCC)[1][3]. PRECSIT expression is repressed by p53, a major tumor suppressor, but is elevated in tumor cells where p53 is dysregulated[1]. Functionally, PRECSIT promotes cancer progression by activating STAT3 signaling and increasing the expression of matrix metalloproteinases, thereby enhancing cell invasion, proliferation, and survival[1][3]. Silencing of PRECSIT or LINC00346 inhibits tumor cell proliferation, migration, and invasion, and induces apoptosis across various cancer types[2][3]. Aberrant expression of PRECSIT is correlated with poorer prognosis, advanced tumor stage, and metastasis in several malignancies[1][3]. No approved drugs directly target PRECSIT, but its role as a pro-oncogenic lncRNA and its regulatory activity over key pathways have made it a candidate for further preclinical investigation as both a prognostic biomarker and a potential therapeutic target[1][3].
