Overview

Long intergenic non-protein coding RNA 355 (LINC00355) is a long non-coding RNA (lncRNA) that does not encode a protein and is primarily involved in the regulation of gene expression at the epigenetic and post-transcriptional levels[3][5]. LINC00355 is upregulated in several cancers, including gastric cancer, lung squamous cell carcinoma, and glioma, where its elevated expression correlates with advanced disease stages and poorer clinical outcomes[1][2][4][6][7]. In cancer, LINC00355 exerts oncogenic functions by acting as a molecular sponge for tumor-suppressive microRNAs, facilitating the degradation of P53 (a critical tumor suppressor) through regulation of specific E3 ubiquitin ligases, and scaffolding epigenetic modifiers to promote the transcription and stability of oncogenic mRNAs (e.g., CDC42)[1][2][4][6][7]. Its subcellular localization is context-dependent, found mainly in the nucleus in gastric cancer cells and in the cytoplasm in glioma and lung squamous cell carcinoma cells[1][2][6]. LINC00355 is under investigation as a prognostic biomarker in multiple cancers and is a candidate for diagnostic and therapeutic targeting, but is not currently a validated therapeutic target for any approved drugs.

Mechanism of action

Acts as a molecular sponge for tumor-suppressive microRNAs (e.g., miR-466 in lung SCC, miR-1225 in glioma). Regulates ubiquitination and degradation of P53 via modulation of E3 ligases RAD18 and UBE3C in gastric cancer. Scaffolds histone acetyltransferases (p300) to promoters and recruits reader proteins for mRNA stabilization (e.g., for CDC42 in gastric cancer).
