Long intergenic non-protein coding RNA 402 (LINC00402) is a conserved long non-coding RNA (lncRNA) gene strongly enriched in T cells and cortical neurons, with higher expression levels noted in human and murine T cells. LINC00402 plays a regulatory role in T-cell responses, specifically augmenting T cell proliferation under allogeneic stimulation, modulating ERK1/2 kinase activity, FOS nuclear localization, and IL-2/Egr-1 expression following T cell receptor engagement, and inversely correlating with T cell activation status. Its expression is influenced by immunosuppressive drugs in transplantation settings, and lower levels are associated with higher risk of acute graft-versus-host disease. LINC00402 additionally participates in ceRNA networks relevant to cancer progression and prognosis. It is a potential therapeutic and prognostic target in immune-related diseases and multiple cancer types, yet direct drug interactions have not been described to date.
