Long intergenic non-protein coding RNA 408 (LINC00408, also known as Lnc-408) is a long non-coding RNA (lncRNA) primarily localized in the cytoplasm and implicated in the regulation of gene expression, particularly in the context of cancer. In breast cancer, LINC00408 acts as a competitive endogenous RNA (ceRNA), sponging miR-654-5p to relieve suppression of LIMK1, thereby promoting epithelial-mesenchymal transition, invasion, metastasis, and cytoskeletal reorganization through the LIMK1/p-cofilin/F-actin axis. Elevated expression of LINC00408 is associated with increased metastatic potential and poor prognosis in breast cancer patients. There is currently no direct evidence for druggability or direct pharmacological targeting, and its main role described in the literature relates to malignancy progression via noncoding regulatory mechanisms[3][5].
