Long intergenic non-protein coding RNA 428 (LINC00428) is a cytoplasmic lincRNA transcript that regulates gene expression and cellular behaviors primarily by acting as a molecular sponge for microRNAs. In breast cancer, LINC00428 (also named Lnc-408 in some literature) sequesters miR-654-5p, relieving microRNA-mediated repression of Lim domain kinase 1 (LIMK1), a protein involved in cytoskeletal actin polymerization and cell invasion. Enhanced expression of LINC00428 promotes invasive and metastatic properties in breast cancer cells, partly by upregulating LIMK1, MMP2, ITGB1, and COL1A1 through the CREB pathway. Its disease association, especially in promoting aggressive cancer phenotypes, makes LINC00428 an attractive but complex target for research into diagnostics and therapeutics.
