Overview

Long intergenic non-protein coding RNA 441 (LINC00441, also known as RB1-DT, ncRNA-RB1) is a divergent long non-coding RNA transcribed in the opposite direction from the RB1 gene locus. It is implicated in the regulation of cell proliferation and oncogenesis primarily through epigenetic silencing of the tumor suppressor RB1 in cancers such as hepatocellular carcinoma, gastric cancer, and cervical cancer. The gene product operates mainly by recruiting DNA methyltransferases to target promoters, thus repressing tumor suppressor gene transcription, and, in some contexts, by sequestering regulatory microRNAs. Aberrant upregulation of LINC00441 is frequently associated with tumor progression. Although not an enzyme, receptor, or classically druggable protein, LINC00441 represents a promising candidate for RNA-based therapeutic strategies and as a biomarker for specific malignancies.
