Overview

Long intergenic non-protein coding RNA 452 (LINC00452) is a human long non-coding RNA (lncRNA). It does not encode a protein but acts as a regulator of gene and protein expression via competitive endogenous RNA activity (microRNA sponging) and by modulating the stability and function of signaling proteins such as ROCK1. LINC00452 is transcriptionally upregulated in ovarian cancer cells and tissues, where it functions to promote tumor cell proliferation, migration, invasion, and aggressiveness, and its elevated expression is associated with poorer prognosis. Mechanistically, LINC00452 acts by sponging tumor-suppressive microRNAs (notably miR-501-3p), thereby stabilizing and increasing the levels of pro-oncogenic effectors such as ROCK1, and also inhibits ROCK1 degradation by the ubiquitin-proteasome system. While primarily studied in ovarian cancer, LINC00452 may have roles in immune cell function and other cancers, but clinical targeting remains in the research phase.

Mechanism of action

Acts as a microRNA sponge, binding and sequestering miR-501-3p, leading to derepression of target mRNAs, prominently ROCK1. Direct interaction with, and inhibition of ubiquitin/proteasome-mediated degradation of, ROCK1 protein, leading to increased Rho/ROCK pathway activity.
