Long intergenic non-protein coding RNA 475 (LINC00475) is a human long non-coding RNA (lncRNA) located at chromosome 9q22.31 (also referred to as C9orf44). LINC00475 does not encode a protein but regulates gene expression by acting as a competing endogenous RNA (ceRNA), binding and sequestering microRNAs such as miR-141-3p and miR-376c-3p. Overexpression of LINC00475 is observed in several cancers, including glioma and papillary thyroid carcinoma, where it promotes cell proliferation, migration, and invasion, correlating with poorer prognosis and shorter survival. In glioma, LINC00475 modulates the Hippo pathway by regulating YAP1 expression through miR-141-3p interaction. These findings highlight LINC00475 as a promising diagnostic and therapeutic target, particularly in oncology.
Acts as a molecular sponge for microRNAs (e.g., miR-141-3p, miR-376c-3p), thereby regulating downstream pathways involved in cell growth and survival. Modulates YAP1 expression (a key regulator in the Hippo pathway) via sequestration of miR-141-3p. Promotes oncogenesis via deregulation of microRNA availability, affecting multiple cancer-related signaling pathways.
