Long intergenic non-protein coding RNA 504 (LINC00504) is a cytoplasm-enriched long non-coding RNA (lncRNA) that plays an important regulatory role in the biology and progression of several cancers, including breast, colon, lung, ovarian, and liver cancers. LINC00504 functions notably as a competing endogenous RNA (ceRNA), where it sequesters specific microRNAs such as miR-876-3p and miR-140-5p, resulting in the upregulation of oncogenic targets like HMGB3 and VEGFA. Overexpression of LINC00504 is commonly associated with increased tumor cell proliferation, migration, invasion, and poor clinical prognosis in several cancer types. Knockdown of LINC00504 induces cancer cell apoptosis, suppresses colony formation and invasion, and reduces tumor growth in vivo. Regulatory transcription factors such as RFX5 can activate LINC00504 transcription. Its disease-association and mechanism suggest that it could serve as a future biomarker for diagnostics or patient stratification and as a potential target for RNA-based therapeutic interventions. There are no drugs directly targeting LINC00504 in clinical use, but experimental strategies involve its inhibition via RNA-targeted therapeutics.
