Long intergenic non-protein coding RNA 518 (LINC00518) is a long non-coding RNA located on chromosome 6p24.3 that is transcribed but not translated to protein[2][3]. LINC00518 is overexpressed in multiple cancers including lung adenocarcinoma, cervical cancer, breast cancer, melanoma, colorectal cancer, prostate cancer, and multiple myeloma[2][5]. It plays critical roles in regulating cell proliferation, apoptosis, migration, invasion, and epithelial-mesenchymal transition, primarily acting as an oncogene by sponging tumor suppressor microRNAs and modulating oncogenic signaling pathways such as Wnt/β-catenin, JAK/STAT3, and HIF-1α[2][3][5]. LINC00518 also confers resistance to radiotherapy and certain chemotherapies by altering hypoxia and glycolysis pathways[3]. High expression of LINC00518 is associated with poor clinical outcomes and advanced disease, making it a promising biomarker for cancer diagnosis and prognosis, as well as a potential therapeutic target, although no LINC00518-directed therapies are yet approved[2][3][5].
