Long intergenic non-protein coding RNA 520 (LINC00520) is a highly conserved long non-coding RNA located on human chromosome 14q22.3[1][3]. It does not encode a protein but instead modulates gene regulation via epigenetic, transcriptional, and post-transcriptional mechanisms. LINC00520 acts as a competing endogenous RNA (ceRNA), binding multiple microRNAs and affecting gene silencing and signaling pathways such as PI3K/AKT and JAK/STAT[1][4]. Aberrant expression of LINC00520 has been observed in various malignancies, including lung, breast, thyroid, colorectal, melanoma, glioma, and other cancers; it generally promotes tumor progression through enhanced proliferation, invasion, and migration, although in cutaneous squamous cell carcinoma (cSCC) it appears to act as a tumor suppressor by downregulating EGFR[1][2]. Its expression levels are associated with prognosis and therapeutic response (e.g., radiosensitivity in head and neck squamous cell carcinoma), highlighting its role as both a potential biomarker and therapeutic target in oncology[1][4]. There are currently no known direct small-molecule or biologic drugs targeting LINC00520.
