Overview

Long intergenic non-protein coding RNA 526 (LINC00526) is a human long non-coding RNA (lncRNA) located on chromosome 18p11.31.[4] It is typically 1322 nucleotides in length and does not encode a protein. LINC00526 is significantly downregulated in glioma and acts as a tumor suppressor by inhibiting glioma cell proliferation, migration, and invasion. Mechanistically, LINC00526 regulates the miR-5581-3p/BEX1 axis as a miRNA sponge, increasing BEX1 expression, and also interacts with EZH2 to repress the transcription of AXL, thereby attenuating the PI3K/Akt/NF-κB signaling pathway. Its reduced expression is linked to poor survival in glioma patients. LINC00526 is being explored as a prognostic biomarker and possible therapeutic candidate for glioma, though it is not considered a traditional drug target such as a receptor, enzyme, or transporter.[1][2][3][4] Note: LINC00526 is not a canonical 'therapeutic target' in the sense of being a receptor, enzyme, or protein targetable by drugs—rather, it is a regulatory lncRNA implicated as a biomarker and a mechanistic contributor to disease.

Mechanism of action

Acts as a competitive endogenous RNA (ceRNA), sponging miR-5581-3p to upregulate BEX1 (brain-expressed X-linked 1) and suppress glioma progression[4] Binds EZH2, represses AXL promoter binding/activation, leading to downregulation of the PI3K/Akt/NF-κB signaling pathway[1][2]
