Long intergenic non-protein coding RNA 543 (LINC00543) is a long non-coding RNA (lncRNA) located in intergenic regions, meaning it does not overlap with protein-coding genes. LINC00543 is overexpressed in certain cancer types, especially colorectal cancer, where its high expression is associated with more advanced disease and poor prognosis. Mechanistically, LINC00543 enhances metastasis by promoting epithelial-mesenchymal transition (EMT) and remodeling the tumor microenvironment. It does this by inhibiting the transport of the precursor microRNA pre-miR-506-3p from the nucleus to the cytoplasm, thereby reducing levels of the mature miR-506-3p. This leads to increased expression of the transcription factor FOXQ1, which induces the expression of CCL2, a chemokine that recruits macrophages and drives them toward an immunosuppressive (M2) phenotype. Through these regulatory processes, LINC00543 contributes to tumor progression and poorer outcomes in patients with colorectal cancer. LINC00543 is not a traditional therapeutic target such as a receptor or enzyme but plays a regulatory role in cancer biology.
