LIN28B-AS1 is a long non-coding RNA that regulates gene expression post-transcriptionally, mainly by stabilizing protein-coding mRNAs (notably LIN28B) through interaction with RNA-binding proteins like IGF2BP1. This lncRNA acts as a "scaffold," facilitating the targeting and stabilizing of oncogenic transcripts and promoting cell proliferation, migration, invasion, and resistance to apoptosis in cancer cells—particularly in hepatocellular carcinoma and lung adenocarcinoma. While not a traditional therapeutic target, LIN28B-AS1 is a key regulatory molecule in the cancer-testis lncRNA family, and holds promise as a biomarker and future therapeutic target for lncRNA-directed approaches.
For potential drugs: Disruption or silencing of LIN28B-AS1 (using siRNA, antisense oligonucleotides, small molecules) could destabilize LIN28B mRNA and reduce oncogenic phenotypes. For indirect effectors: Curcumin attenuates chemoresistance by inhibiting NF-κB-induced LIN28B expression.
