Long intergenic non-protein coding RNA 601 (LINC00601) is a long non-coding RNA located on chromosome 10q26.2 and transcribed from the minus strand, spanning approximately 8 kb. LINC00601 is not translated into protein but acts as a regulatory RNA molecule involved in cancer biology. Elevated expression of LINC00601 has been observed in multiple malignancies, including gliomas and hepatocellular carcinoma, where it is associated with tumor grade, increased cellular proliferation, migration, invasion, and poorer prognosis. In glioma, LINC00601 modulates tumor biology mainly by regulating the JAK/STAT (specifically p-STAT3) signaling pathway, and possibly interacts with additional oncogenic axes such as the NOD-like receptor and TNF pathways. Its silencing leads to reduced tumor cell proliferation, invasion, and in vivo tumor growth. LINC00601 is thus considered a potential prognostic biomarker and a candidate molecular target for therapeutic intervention in selected cancers. No approved drugs directly target LINC00601 in clinical practice, but research has explored combined targeting with STAT3 inhibitors such as Stattic. LINC00601 is a long non-coding RNA functional in cancer signaling, considered a promising but research-stage therapeutic biomarker and target, especially in glioma and liver cancer. Its regulation of JAK/STAT—particularly p-STAT3—links it to oncogenic proliferation and survival pathways. There are currently no established, specific drug interactions, but experimental studies demonstrate the potential for combinatorial inhibition strategies with agents like Stattic.
LINC00601 modulates the JAK/STAT signaling pathway (notably p-STAT3 in glioma) and exhibits potential competitive endogenous RNA (ceRNA) activity to sequester miRNAs affecting oncogenic signaling.
