Overview

LINC00607 (long intergenic non-protein coding RNA 607) is a human-specific long non-coding RNA expressed from chromosome 2q35[3]. It is labeled as an lncRNA (long non-coding RNA) and does not encode a protein. LINC00607 is found in the cytoplasm and nucleus, where it regulates diverse cellular processes. In cancer, particularly osteosarcoma, LINC00607 promotes cell proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) by acting as a sponge for miR-607, thereby enhancing E2F6 protein expression and downstream oncogenic pathways[1][4]. In vascular endothelial and smooth muscle cells, it modulates angiogenesis, extracellular matrix remodeling, inflammatory responses, and cell homeostasis; these actions are mediated both in cis (local regulation of nearby genes like FN1) and in trans (broad control of gene expression networks), including as part of c-Myc-dependent regulatory circuits[2]. LINC00607 has been identified as an upregulated target in various pathological states, making it a promising but challenging candidate for therapeutic intervention and as a disease biomarker. At present, no drugs directly target LINC00607, but antisense or RNA-targeted drug development is under research consideration.

Mechanism of action

Not directly druggable at present; however, LINC00607 acts as: - Competing endogenous RNA (ceRNA) by sponging miR-607, thereby upregulating E2F6 protein expression[1][4] - Regulator of c-Myc-target gene expression networks[2] - Possible future mechanisms include antisense oligonucleotides or other RNA-targeted approaches if therapeutically developed.

Safety considerations

Therapeutic targeting of lncRNAs may cause off-target effects or disrupt essential regulatory networks in normal tissues[1][2].LINC00607 is human-specific (lacks orthologs in many model organisms), which may pose translational challenges for preclinical safety studies[1].Modulating LINC00607 may impact processes such as vascular homeostasis and tissue repair, with unclear systemic consequences[2].
06

Biomarkers

High expression of LINC00607 is correlated with increased proliferation and invasiveness in osteosarcoma[1][4].Elevated LINC00607 expression marks endothelial cell dysfunction and vascular remodeling in cardiovascular disease models[2].Potential biomarker for patient selection and prognosis in certain cancers[1].
