Long intergenic non-protein coding RNA 612 (LINC00612) is a human long non-coding RNA transcribed from a locus on chromosome 12, also known as C12orf33. This lncRNA does not encode a protein but exerts regulatory functions in gene expression networks. LINC00612 acts both in the cytoplasm, serving as a competitive endogenous RNA (ceRNA) by sponging specific microRNAs, and in the nucleus by recruiting transcription factors (like phosphorylated STAT3) to gene promoters, notably regulating genes such as alpha-2-macroglobulin (A2M). In the context of disease, LINC00612 is implicated in multiple cancers where it promotes cellular proliferation and invasion, and in COPD, where its downregulation is associated with increased apoptosis and inflammation. Although it is not a classical therapeutic target such as a receptor or enzyme, LINC00612 is an emerging regulatory RNA with potential as a biomarker and a candidate for RNA-targeted therapeutic strategies[1][2][3][4][6][8][10].
Acts through molecular sponging of microRNAs (such as miR-214-5p, miR-590-3p, miR-31-5p), and recruitment of transcription factors such as STAT3 to gene promoters influencing gene expression and phenotypic outcomes[2][4][6][10]
