Overview

Long intergenic non-protein coding RNA 621 (LINC00621) is a human lncRNA gene found on chromosome 13q12.12[15]. It encodes a long non-coding RNA molecule variably expressed in normal and tumor tissues, notably upregulated in lung adenocarcinoma and associated with poor clinical outcomes[2]. LINC00621 acts mechanistically as a molecular sponge for miR-34a-5p, resulting in the upregulation of TGFBR1 and increased activation of the TGF-β signaling pathway, promoting tumorigenesis and metastasis[2]. Its expression is transcriptionally upregulated by FOXA1, a transcription factor involved in chromatin remodeling. LINC00621 is being actively investigated as a biomarker for cancer prognosis and as a potential therapeutic target[2][5][6][15].

Mechanism of action

As of current data, drugs would be expected to modulate LINC00621 expression or function, likely through antisense oligonucleotides, RNA interference, or small molecules influencing its transcriptional regulation or miRNA sponging activity[2].

03

Biological functions

Regulation of gene expression and chromatin remodelingModulation of cell proliferation, migration, and invasion (esp. in lung adenocarcinoma)Functions as a molecular sponge for specific microRNAs, influencing downstream gene activity (e.g., miR-34a-5p/TGFBR1/TGF-β axis)
04

Disease associations

Cancer (especially lung adenocarcinoma and possibly other malignancies)Possible roles in mucopolysaccharidosis type II and limb-girdle muscular dystrophy (less established)
05

Safety considerations

Potential off-target effects and toxicity associated with RNA-targeted therapies.Unknown risks related to modulating lncRNA expression, given the broad regulatory roles of these molecules in normal cell physiology[2].
06

Biomarkers

LINC00621 expression levels in tumor tissue (e.g., elevated in lung adenocarcinoma, associated with poor prognosis)[2]miR-34a-5p levels (inverse relationship with LINC00621)[2]
