Long intergenic non-protein coding RNA 623 (LINC00623) is a long non-coding RNA that does not code for a protein but plays important regulatory roles in cancer biology, particularly in pancreatic ductal adenocarcinoma (PDAC)[1][2][3]. LINC00623 is upregulated in PDAC tissues and exosomes, promotes tumor cell proliferation, migration, and invasion, and is associated with poor patient outcomes[1][2]. Mechanistically, LINC00623 acts in the nucleus as a scaffold, recruiting the deubiquitinase USP39 to N-acetyltransferase 10 (NAT10), preventing NAT10 ubiquitination and proteasomal degradation, which in turn stabilizes mRNAs with oncogenic potential through enhanced N4-acetylcytidine (ac4C) modification. Enhanced LINC00623 expression serves as a promising diagnostic and prognostic biomarker, and the LINC00623/NAT10 axis is a potential therapeutic target for PDAC, although no drugs currently target this RNA directly[1][2][3][6].
