Long intergenic non-protein coding RNA 628 (LINC00628) is a long non-coding RNA implicated in cancer. It can function either as a tumor suppressor or an oncogenic factor, depending on the tissue context. For example, in *gastric cancer*, upregulation of LINC00628 suppresses tumor growth, migration, and invasion, in part by modulating histone modifications and interacting with polycomb repressive complex 2 (EZH2). In *lung adenocarcinoma*, LINC00628 is overexpressed and associated with poor survival—suggesting it may have an oncogenic role and potential as a prognostic biomarker. LINC00628 regulates gene expression epigenetically, in part by recruiting DNA methyltransferases (DNMT1, DNMT3A, DNMT3B) or histone methyltransferase activities to target gene promoters such as LAMA3, thereby modifying methylation-dependent gene silencing. Currently, there are no approved drugs directly targeting LINC00628, but its unique expression profile in cancers makes it a candidate for future RNA-based or epigenetic therapies.
