Overview

Long intergenic non-protein coding RNA 629 (LINC00629) is a long non-coding RNA mapped to chromosome X (Xq26) and is most highly expressed in the placenta and reproductive tissues. It has several isoforms (LINC00629A, LINC00629B, LINC00629C, and LINC00629D) differing in exon structure[5]. LINC00629 is implicated in the regulation of gene expression via chromatin modification and mRNA stability, and its subcellular localization indicates both nuclear and cytoplasmic functions[5]. Experimental data demonstrate that certain LINC00629 isoforms suppress migration and invasion of choriocarcinoma cells, suggesting a tumor suppressor role; its downregulation is associated with increased aggressiveness[5]. In cancer, its expression is often altered: LINC00629 is downregulated in ovarian and gastric cancer, and its overexpression can inhibit glycolysis (by destabilizing c-Myc), proliferation, and metastasis in ovarian and gastric cancer models[3][4]. LINC00629 acts as a molecular sponge for miRNAs (e.g., miR-196b-5p), leading to the upregulation of targets such as AQP4 and participating in complex regulatory networks[4]. There are no known drugs targeting LINC00629, and its primary utility is as a potential prognostic tumor biomarker or mechanistic regulator rather than a direct therapeutic target[2][3][4][5].
