Overview

Long intergenic non-protein coding RNA 630 (LINC00630) is a long non-coding RNA gene located on chromosome Xq22.1, does not code for protein, and is primarily detected in the cytoplasm and nucleus[1][3][5]. LINC00630 is upregulated in several cancer types, such as cholangiocarcinoma and hepatocellular carcinoma, and contributes to tumor progression by promoting cell proliferation, migration, invasion, and by inhibiting apoptosis. Mechanistically, it acts as a competing endogenous RNA (ceRNA), sponging microRNA-199a to regulate the expression of fibroblast growth factor 7 (FGF7), and can recruit the transcription factor E2F1 to the CDK2 promoter to increase CDK2 transcription, affecting cell cycle progression[1][2][4][6]. Its expression correlates with advanced disease and poor prognosis, making it a potential therapeutic target and biomarker in oncology. No clinically approved drugs directly target LINC00630 currently.

Mechanism of action

Modulation of microRNA sponging (e.g., miR-199a sponging to upregulate FGF7); Recruitment of transcription factors (e.g., E2F1 to the CDK2 promoter); Epigenetic modulation (potential in targeting regulatory axes such as BEX1/EZH2/CDK2)
