Long intergenic non-protein coding RNA 653 (LINC00653), also known as lysosome cell death regulator (LCDR), is a nuclear long non-coding RNA (lncRNA) highly expressed in certain cancer cell lines (notably p53-null non–small-cell lung cancer cells)[1]. LCDR regulates the integrity of the lysosomal membrane by stabilizing the LAPTM5 mRNA transcript through interaction with the RNA-binding protein hnRNPK, thereby promoting cell survival and preventing lysosomal cell death. Through this mechanism, LCDR influences lysosome-mediated cell death pathways and affects cell proliferation, colony formation, and tumorigenesis, at least in experimental models. Although LCDR is being explored for its role in cancer and its potential as a biomarker, it is not regarded as a classical therapeutic target like a receptor, enzyme, or transporter, but rather as a regulatory RNA implicated in disease biology[1][7]. Notes and caveats: - LCDR/LINC00653 is an lncRNA and not a protein-coding gene, enzyme, or receptor; therefore, it is not conventionally considered a “target” for small molecule drugs or biologics in the same sense as receptors or enzymes[1][4][7]. - No known drugs directly target LCDR/LINC00653, nor are there validated mechanisms of action or clinical interventions based on this RNA at present[1][7]. - In the literature, LCDR is a research term rather than a widely adopted gene/protein symbol, and the biology, while promising, is incompletely characterized[1]. - LCDR and LINC00653 refer to the same lncRNA locus; the “lysosome cell death regulator” designation is based on functional studies in selected cancer cells, not a universal annotation[1][7]. - The field of lncRNAs is emerging, and information about their therapeutic relevance is subject to change as new research is published[1][4].
