Long intergenic non-protein coding RNA 654 (LINC00654) is a long non-coding RNA (lncRNA) gene located on chromosome 20p12.3, also known as LINC01729 or ENSG00000205181[1][3]. As a member of the lncRNA class, LINC00654 does not code for a protein but is involved in the regulation of gene expression networks and interacts with various mRNA targets. LINC00654 functions as a “hub lncRNA” and is implicated in the regulation of cell signaling pathways including GTPase signaling, Notch and Hedgehog signaling, cell cycle, immune modulation, and tissue homeostasis[2][3][6][7]. It is upregulated in several cancers, such as breast, colorectal, ovarian cancer, and diffuse large B-cell lymphoma, where it frequently acts as a risk or prognostic factor[2][7]. Outside cancer, LINC00654 is associated with the regulation of extracellular matrix and chondrocyte homeostasis (in osteoarthritis) and is differentially expressed during spermatogenesis[7]. LINC00654 is not currently a direct therapeutic target (no drugs known to act on it directly), but its roles as a biomarker and regulator of disease-relevant pathways make it of growing biomedical interest.
