Overview

Long intergenic non-protein coding RNA 674 (LINC00674), also known as CALA (cytoplasmic G3BP1-associated lncRNA), is a spliced multi-exonic long non-coding RNA gene located on human chromosome 17q24.2. It does not encode a protein but plays a key role as a cytoplasmic regulator of gene expression, primarily by scaffolding ribonucleoprotein complexes involving G3BP1, G3BP2, UPF1, and associated NMD factors[2][13]. CALA modulates RNA turnover rates and the efficiency of nonsense-mediated mRNA decay in endothelial cells, thereby affecting cellular homeostasis, migration, and stress responses[2][4][6][10]. Although not a conventional drug target, its biological functions are implicated in endothelial physiology and cancer, making it a subject of ongoing molecular research. There are no known drugs directly interacting with LINC00674, and its clinical therapeutic utility remains unexplored[2][11][13].

Mechanism of action

Not applicable. No approved or investigational drugs have a defined mechanism of action involving LINC00674

03

Biological functions

Regulation of RNA turnover, particularly nonsense-mediated mRNA decay (NMD) in the cytoplasmScaffold for cytoplasmic ribonucleoprotein (RNP) complexes, notably those including G3BP1, UPF1, and partnersRegulation of endothelial cell function and possibly cell migration and sproutingModulation of gene expression post-transcriptionallyPotential role in stress response and homeostasis
04

Disease associations

Cancer (reported expression in diverse cancer tissues, potential relevance via NMD regulation)Endothelial function/dysfunction (possibly cardiovascular disease roles)Other (potential in RNA metabolism disorders, still under investigation)
05

Safety considerations

None specific documented for therapeutic manipulation, as this RNA is not a clinical drug target. This area would require further investigation if therapeutic modulation were considered
06

Biomarkers

Expression of CALA/LINC00674 (itself) as a biomarker—for cancer and possibly endothelial stress, pending further validationLevels of known NMD targets (indirectly, as CALA affects NMD pathway activity and gene expression profiles in endothelial cells)
