Overview

Long intergenic non-protein coding RNA 702 (LINC00702) is a long non-coding RNA (lncRNA) that does not encode a protein but regulates cellular processes via RNA-based mechanisms. It is notably downregulated in several cancers, including non-small cell lung cancer and gastric cancer, where lower levels are associated with poorer prognosis features such as increased tumor size and metastasis. Mechanistically, LINC00702 acts as a competing endogenous RNA (ceRNA) that sponges miR-510, relieving suppression of the tumor suppressor PTEN and downregulating PI3K-Akt signaling, resulting in suppressed tumor growth, invasion, and increased apoptosis. LINC00702 is of increasing interest as a diagnostic/prognostic biomarker and a potential therapeutic target in oncology

Other names
RP11-67B16.1
02

Mechanism of action

Functions as a ceRNA for miR-510, thereby upregulating the tumor suppressor gene PTEN, inhibiting the PI3K-Akt pathway, and suppressing tumor proliferation and metastasis

03

Biological functions

Regulation of cell proliferationRegulation of apoptosisCompeting endogenous RNA (ceRNA) activityRegulation of gene expression (notably modulation of PTEN via miR-510 sponge activity in cancer models)
04

Disease associations

Cancer (notably non-small cell lung cancer, gastric cancer, high-grade serous ovarian cancer)
05

Safety considerations

No specific therapeutic safety issues reported, but general challenges include delivery and off-target effects inherent to RNA-based or gene therapy approaches
06

Interacting drugs

None directly reported (lncRNAs are not directly targeted by small-molecule drugs as of current knowledge, but could be manipulated via nucleic acid therapeutics or indirectly through gene therapy strategies)
07

Biomarkers

Candidate diagnostic and prognostic biomarker in non-small cell lung cancer and possibly gastric and ovarian cancers based on altered expression in tumors vs. normal tissue
