Long intergenic non-protein coding RNA 842 (LINC00842) is a long non-coding RNA molecule that has no protein-coding potential and mainly functions as a regulatory RNA. It is upregulated in pancreatic ductal adenocarcinoma (PDAC) and drives malignant cellular phenotypes including proliferation, migration, invasion, and metabolic reprogramming. Mechanistically, LINC00842 interacts with the transcriptional co-activator PGC-1α, preventing its deacetylation by SIRT1, which leads to a metabolic shift favoring fatty acid synthesis over mitochondrial oxidative metabolism. High levels of LINC00842 and acetylated PGC-1α are correlated with poor patient survival in PDAC. LINC00842 is considered a potential therapeutic target, and silencing it or inhibiting its downstream metabolic effectors suppresses tumor progression in preclinical models. It appears to have tissue-specific functions that may also play roles in other cancer types
For experimental interventions: RNA interference (RNAi) or antisense oligonucleotides (ASOs) targeting LINC00842 decrease tumor growth, proliferation, migration, and invasion; Inhibition of downstream metabolic targets (e.g., fatty acid synthase) represses tumor growth when LINC00842 levels are high; Mechanistic action: Disruption of LINC00842 binding to PGC-1α alters its acetylation state and metabolic gene programs
