Overview

Long intergenic non-protein coding RNA 844 (LINC00844) is a long noncoding RNA predominantly localized in the cytoplasm, enriched especially in the liver, brain, and prostate[2]. It regulates the expression of key drug metabolizing enzymes (CYP3A4, CYP2E1, SULT2A1) and nuclear receptors (PXR, HNF4α) by acting as a molecular sponge for specific microRNAs (notably hsa-miR-486-5p) and by recruiting transcription factors to gene promoters[2][1]. LINC00844 has been shown to suppress proliferation and promote apoptosis in prostate cancer cells by upregulating GSTP1 through the recruitment of EBF1 to the GSTP1 promoter[1]. In hepatocellular carcinoma, its expression is downregulated, and overexpression can inhibit tumor cell proliferation and invasion, indicating a tumor-suppressive role[4]. LINC00844 expression is responsive to drug-induced liver toxicity (e.g., acetaminophen exposure), and its regulatory network may influence susceptibility to drug-induced adverse effects[2]. Although not directly targeted by therapeutic drugs, it is considered a potential therapeutic target and biomarker in oncology and pharmacogenomics research[1][2][4].

Mechanism of action

Not directly drug-targeted; modulates cellular processes through regulation of drug metabolizing enzymes (CYP3A4, CYP2E1, SULT2A1) and nuclear receptors (PXR, HNF4α) via miRNA and protein interactions[2].
