Overview

Long intergenic non-protein coding RNA 853 (LINC00853) is a human long non-coding RNA, 1826 nucleotides in length, located on chromosome 1p33. While not encoding a protein, LINC00853 plays significant regulatory roles in cancer biology. Recent studies have shown it is upregulated in hepatocellular carcinoma (HCC) and gastric cancer, particularly in extracellular vesicle (exosome) fractions from patient serum or cell lines. In HCC, exosomal LINC00853 is an effective and noninvasive biomarker, displaying superior diagnostic performance over traditional alpha-fetoprotein (AFP), including cases where AFP is negative. In gastric cancer, LINC00853 promotes cell proliferation, migration, and invasion, partly through activation of the MAP17/PDZK1/AKT pathway, and its exosomal form can serve as a biomarker for disease occurrence and recurrence. In T-cell acute lymphoblastic leukemia, downregulation of LINC00853 is associated with increased cell migration and invasion, suggesting a context-dependent tumor suppressor role. No approved drugs are known to directly target LINC00853, and current clinical application is limited to its use as a disease biomarker.

Mechanism of action

Acts as an oncogenic lncRNA by activating the MAP17/PDZK1/AKT signaling pathway in gastric cancer. Possibly regulates signaling pathways and gene expression via epigenetic modification, miRNA sponging, or interaction with chromatin/proteins (general lncRNA mechanisms, inferred from context).
