Long intergenic non-protein coding RNA 858 (LINC00858) is a long non-coding RNA molecule, approximately 2685 nucleotides in length, located at 10q23.1. LINC00858 exhibits abnormally high expression in a wide range of human cancers, notably colorectal cancer and non-small cell lung cancer, where it is associated with tumor progression, poor prognosis, and increased metastatic potential. Mechanistically, LINC00858 acts as a competitive endogenous RNA (ceRNA), sponging various microRNAs such as miR-25-3p, miR-22-3p, miR-3182, and miR-422a, thereby modulating downstream gene targets including SMAD7, YWHAZ, KLK4, and CDK14. In colon cancer, LINC00858 recruits the transcription factor RAD21 to the promoter of the PCNP gene, activating the STAT3/5 pathway. Its dysregulation promotes cell proliferation, migration, invasion, suppression of apoptosis, and is tied to clinical features of tumor aggressiveness and advanced stage. LINC00858 is regarded as a novel oncogenic lncRNA and emerging therapeutic target, although no current drugs directly target this molecule. Detection of LINC00858 expression is increasingly recognized as a valuable biomarker for identifying high-risk cancer patients and monitoring disease progression.
