Overview

Long intergenic non-protein coding RNA 880 (LINC00880) is a long non-coding RNA characterized by its lack of protein-coding potential. It acts as an oncogenic lncRNA in several cancer types, most notably colorectal cancer and lung adenocarcinoma. In colorectal cancer, LINC00880 is overexpressed and correlates with poor patient prognosis, promoting cell proliferation and tumor growth through transcriptional activation of the MCM3 gene and modulation of cell cycle and DNA replication pathways. In lung adenocarcinoma, LINC00880 is regulated by super-enhancers and the transcription factor FOXP3, acting as a structural scaffold for the formation of a ternary complex between CDK1 and PRDX1. This complex increases CDK1 kinase activity, influencing the PI3K/AKT signaling pathway and sustaining malignancy. LINC00880 has emerged as a potential diagnostic and prognostic biomarker as well as a therapeutic target in cancer, but has yet to be targeted by approved therapies. Its levels may modulate sensitivity to DNA replication or helicase inhibitors and to epigenetic drugs that target super-enhancers.

Mechanism of action

Inhibition of LINC00880 reduces cell proliferation and tumor growth via suppression of cell cycle genes (e.g., MCM3) and DNA replication pathways. Drugs targeting epigenetic regulation of super-enhancers suppress LINC00880 transcription.
