Overview

Long intergenic non-protein coding RNA 891 (LINC00891) is a long non-coding RNA transcribed from chromosome X (chrX:71697196-71706455). It is a non-protein coding gene, confirmed by multiple computational and experimental assessments. LINC00891 is downregulated in lung cancer tissues and functions as a tumor suppressor by inhibiting cell proliferation, migration, invasion, and the epithelial-to-mesenchymal transition (EMT) process. Mechanistically, it suppresses the RhoA pathway and is transcriptionally regulated by GATA2, which in turn is regulated by miR-128-3p. LINC00891 knockdown promotes malignant progression and xenograft tumor growth, whereas its overexpression is associated with reduced malignancy in preclinical models. CCG-1423, an inhibitor of the RhoA pathway, can antagonize the pro-tumorigenic effects resulting from LINC00891 knockdown. The molecule is considered a promising avenue for therapeutic intervention and as a prognostic biomarker in lung cancer. There are currently no approved drugs targeting LINC00891 directly nor known safety concerns, but RNA-targeting strategies remain experimentally challenging.
