Overview

Long intergenic non-protein coding RNA 899 (LINC00899) is a multi-exonic, chromatin-associated long non-coding RNA located on chromosome 22q13.31. It serves as a multifunctional regulator with context-dependent roles in human disease, primarily modulating cellular processes via acting as a competing endogenous RNA (ceRNA) that sponges microRNAs such as miR-944, miR-425, and miR-744-3p. In several cancers, including cervical cancer, breast cancer, spinal ependymoma, and acute myeloid leukemia, LINC00899 regulates cell proliferation, viability, migration, and invasion by modulating the expression of downstream targets (e.g., ESR1, DICER1, YY1, RUNX2, and TPPP/p25), primarily through miRNA-dependent and direct chromatin-associated transcriptional mechanisms. Elevated LINC00899 is associated with better prognosis in breast cancer but correlates with poor outcomes in some hematologic malignancies, such as AML. Beyond oncology, LINC00899 promotes osteogenic differentiation and may be implicated in osteoporosis prevention. Despite disease relevance, there are currently no known drugs that directly target LINC00899, and it is not classified as a conventional therapeutic target such as a receptor or enzyme[1][2][3].
