Overview

Long intergenic non-protein coding RNA 908 (LINC00908) is a long non-coding RNA located on chromosome 18; it encodes a 60-amino-acid regulatory peptide known as ASRPS (a small regulatory peptide of STAT3). LINC00908 and its encoded micropeptide are downregulated in triple-negative breast cancer (TNBC) and other cancers, correlating with increased tumor growth and poor prognosis. ASRPS directly binds to the coiled-coil domain of STAT3, inhibiting STAT3 phosphorylation and subsequent VEGF expression, thereby suppressing tumor angiogenesis. LINC00908 is transcriptionally regulated by estrogen receptor alpha (ERα), and its expression appears to be negatively regulated in ERα-negative tumors such as TNBC. There is emerging evidence for its potential as a therapeutic target and prognostic cancer biomarker, but no drugs are currently known to target this lncRNA or its peptide directly[1][2][3][4].

Mechanism of action

Encodes the 60-amino acid peptide ASRPS, which binds to the coiled-coil domain of STAT3, inhibiting its phosphorylation and downstream VEGF expression, resulting in decreased tumor angiogenesis[1][2][3][4].

Disease associations

Cancer (with particular focus on triple-negative breast cancer)Potential involvement in liver cancer and prostate cancer
05

Safety considerations

No specific safety concerns or therapeutic challenges reported to date.
06

Biomarkers

Downregulation of LINC00908 or ASRPS associated with poor overall survival in triple-negative breast cancer[1][2]. Expression of ASRPS may serve as a prognostic biomarker[1][2].
