Overview

Long intergenic non-protein coding RNA 909 (LINC00909, also known as ZNF407 antisense RNA 1 or ZNF407-AS1) is a cytoplasmic long non-coding RNA transcribed from the antisense strand of the ZNF407 gene locus. It does not code for a protein and instead acts primarily through post-transcriptional mechanisms, notably by destabilizing SMAD4 mRNA, leading to downregulation of SMAD4 and subsequent upregulation of pluripotency genes and activation of oncogenic signaling pathways (e.g., MAPK/JNK). LINC00909 is overexpressed in several cancers, including pancreatic ductal adenocarcinoma and glioma, where it promotes cancer stemness, tumor metastasis, and is associated with poor clinical outcomes. Its functional role as a regulator of mRNA stability and signaling makes it of interest as a biomarker and a potential (though not yet directly druggable) molecular target in cancer[1][2][4].
