Overview

Long intergenic non-protein coding RNA 930 (LINC00930) is a long non-coding RNA molecule, mainly nuclear, without protein-coding potential[1][2]. It functions primarily as an epigenetic regulatory scaffold, facilitating the interaction of histone-modifying enzymes (RBBP5 and GCN5) to modify chromatin states and activate transcription of metabolic and proliferative genes, especially PFKFB3, in nasopharyngeal carcinoma and other cancer types[2]. LINC00930 promotes tumor cell proliferation and glycolysis by enhancing histone H3 lysine 4 trimethylation and H3 lysine 9 acetylation at specific gene promoters[2]. Dysregulation of LINC00930 is associated with tumorigenesis, altered metabolism, and poor prognosis in several cancers, including nasopharyngeal carcinoma (where it is typically upregulated and oncogenic) and pancreatic cancer (where it may act as a tumor suppressor)[2][3]. LINC00930 is being investigated as a potential biomarker and therapeutic target for cancer, although no approved drugs currently directly target this lncRNA. Drugs that block downstream effectors in its pathway (e.g., OICR-9429, YF-2) can affect its functional output in preclinical studies[2].

Mechanism of action

Epigenetic modulation via scaffolding RBBP5/GCN5, leading to histone methylation/acetylation of target promoters (notably PFKFB3); Indirect modulation of metabolic enzymes and cell proliferation regulators; Competing endogenous RNA activity (in some cancer contexts, by sponging miRNAs).
