Overview

Long intergenic non-protein coding RNA 945 (LINC00945) is a human long intergenic non-coding RNA (lincRNA) that does not encode protein but regulates gene expression at multiple levels, particularly through epigenetic and chromatin-associated mechanisms. It is identified as a super-enhancer-associated lncRNA (SE-lncRNA) in glioma, where it is highly upregulated compared to normal brain tissue. Elevated LINC00945 expression promotes tumor cell proliferation, epithelial-mesenchymal transition, migration, invasion, and in vivo tumor growth, contributing to glioma pathogenesis and poor prognosis. Mechanistically, its expression is regulated by super-enhancers involving cofactors such as BRD4 and MED1, which are key chromatin regulators implicated in cancer. LINC00945 is under study as a prognostic biomarker and is considered a novel candidate therapeutic target in glioma research. Its biological functions, however, are not limited to cancer and may extend to the fine-tuning of gene expression across various tissues, typical of lincRNAs [1][2][5].

Mechanism of action

Not established; inferred mechanisms would involve epigenetic regulation or modulation of transcriptional networks due to its effect on chromatin structure and gene transcription

03

Biological functions

Regulation of gene expression (especially via epigenetic and chromatin interactions)Promotion of glioma cell proliferationEnhancement of epithelial-mesenchymal transition (EMT), migration, and invasion in glioma cellsPotential scaffolding and modulation of protein activities
04

Disease associations

Cancer, notably glioma (central nervous system tumors)Possible role in prognosis and therapy resistance in glioma
05

Safety considerations

Unknown for direct targeting, as no drugs are reported to target LINC00945 specifically. General safety concerns for lncRNA-targeting therapeutics may include effects on gene regulation networks and tissue specificity
06

Interacting drugs

None reported as directly targeting LINC00945 as of the available literature.
07

Biomarkers

High LINC00945 expression is a biomarker for poor prognosis in gliomaPart of a multi-lncRNA risk signature predictive of glioma patient outcome and immunotherapy response
