Long intergenic non-protein coding RNA 955 (LINC00955) is a long non-coding RNA (lncRNA) found in the human genome (chromosome 4p16.3, ~2483 nucleotides in length) that does not encode a protein but acts at the RNA level to regulate cellular processes. Recent studies show that LINC00955 acts as a scaffold molecule, facilitating the interaction between proteins such as the E3 ubiquitin ligase TRIM25 and the transcription factor Sp1, which leads to post-translational regulation and degradation of Sp1. This, in turn, inhibits DNA methyltransferase 3 beta (DNMT3B) expression and results in hypomethylation of the PHIP promoter, inducing PHIP expression and degradation of cyclin-dependent kinase 2 (CDK2), ultimately causing cell cycle arrest in G0/G1 and suppression of colorectal cancer cell growth. Lower levels of LINC00955 are correlated with poorer prognosis in colorectal cancer, and it has been suggested as a potential therapeutic target and prognostic biomarker in this context. No direct drug interactions or targeting mechanisms or notable safety concerns have been reported for LINC00955.
