Overview

Long intergenic non-protein coding RNA 992 (LINC00992), also called ERINA (estrogen inducible lncRNA), is a long non-coding RNA (lncRNA) encoded on chromosome 5 (5q23.1) that does not code for a protein but acts as a regulatory RNA[1][3]. LINC00992/ERINA is highly expressed in estrogen receptor (ER)-positive breast cancers and its expression is associated with poor prognosis and increased resistance to endocrine therapies and CDK inhibitors among affected patients[2][4][5]. Mechanistically, LINC00992/ERINA drives cell cycle progression and proliferation by interacting with the transcription factor E2F1, preventing its inhibition by the tumor suppressor RB1[2][4]. LINC00992 expression is responsive to estrogen; estrogen or ER agonists increase its expression, while anti-estrogen agents like tamoxifen suppress it[2][5]. Experimentally, altering LINC00992 levels modulates response to tamoxifen and affects cellular viability and migration, suggesting a functional role in endocrine resistance[5]. Because of its strong association with prognosis and treatment response in ER-positive breast cancer, LINC00992/ERINA is under investigation as a prognostic and predictive biomarker, and potential therapeutic target, although no specific drugs directly and selectively target this lncRNA in clinical practice yet[2][5].
