Overview

Long intergenic non-protein coding RNA 994 (LINC00994) is a long non-coding RNA (lncRNA) gene that does not encode a protein but exerts regulatory effects in various cancer types, notably pancreatic and gastric cancer[1][2][3][4]. It is upregulated in tumor samples and cell lines, promoting proliferation, migration, invasion, and cell cycle progression, while inhibition leads to cell cycle arrest (G1 phase) and apoptosis[2][3][4]. Mechanistically, LINC00994 acts as a competing endogenous RNA, sponging microRNAs such as miR-765-3p, which in turn modulates downstream targets like Runt-related transcription factor 2 (RUNX2)[2][4]. This regulatory network contributes to the aggressive phenotype of cancer cells, and silencing LINC00994 impairs tumor cell growth and invasiveness[2][3][4]. Because of its central role in cancer biology, LINC00994 is considered a potential therapeutic target and biomarker, but no interacting drugs or therapies have been reported to directly target it as of the current literature[2][3][4].
