Overview

Long Interspersed Nuclear Element-1 (LINE-1) Open Reading Frame 1 protein (ORF1p) is a 40 kDa RNA-binding protein and nucleic acid chaperone essential for the mobilization of LINE-1 retrotransposons (UniProt P12287). While LINE-1 elements are typically silenced in healthy adult somatic tissues through epigenetic mechanisms, they are frequently derepressed in a wide variety of cancers, including breast, lung, and colorectal carcinomas, where ORF1p expression serves as a highly specific biomarker (Taylor et al., 2023, Nature). ORF1p binds to LINE-1 mRNA to form a ribonucleoprotein (RNP) complex, facilitating its transport to the nucleus for reverse transcription and integration, a process that promotes genomic instability and triggers innate immune responses via the cGAS-STING pathway (De Cecco et al., 2019, Nature). Therapeutic strategies targeting ORF1p include small molecule inhibitors of its chaperone activity and immunotherapeutic approaches, such as vaccines or antibodies, aimed at selectively eliminating ORF1p-expressing malignant cells. Given its near-absence in normal tissues and high prevalence in tumors, ORF1p represents a promising pan-cancer target for both diagnostic and therapeutic applications.
