Long non-coding RNA 34a host gene (MIR34AHG), also known as lnc34a, is a long non-coding RNA enriched in colon cancer stem cells and initiates asymmetric division by targeting the microRNA miR-34a for spatially imbalanced epigenetic silencing. lnc34a recruits the DNA methyltransferase Dnmt3a—via PHB2—and HDAC1, leading to methylation and deacetylation of the miR-34a promoter, resulting in its silencing. Its upregulation is associated with late-stage colorectal cancer, contributing to cancer stem cell self-renewal and tumor proliferation. MIR34AHG itself is not a receptor, enzyme, transporter, or classical therapeutic target; rather, it is a regulatory lncRNA influencing the tumor suppressor microRNA miR-34a and thus plays a crucial role in cancer biology[1].
Not applicable as a direct drug target; however, its suppression leads to increased miR-34a expression, which is tumor-suppressive[1].
