Long non-coding RNA activating regulator of DKK1 (LNCAROD) is a long non-coding RNA (lncRNA) located in the nucleus with regulatory roles in gene expression, notably acting as an enhancer RNA that upregulates DKK1 expression through chromatin remodeling and recruitment of regulatory proteins[2]. LNCAROD is stabilized by m6A methylation and plays an oncogenic role in cancers such as head and neck squamous cell carcinoma (HNSCC), where it scaffolds an interaction between HSPA1A and the transcription factor YBX1, protecting YBX1 from proteasomal degradation and thus promoting cell proliferation and tumor progression[3]. LNCAROD also acts as a competitive endogenous RNA (ceRNA), binding to microRNAs like miR-324-5p and influencing downstream immune regulatory pathways, such as the CUEDC2 axis in the antiviral response[4]. Overexpression of LNCAROD is associated with adverse outcomes in multiple cancers. Due to its diverse regulatory mechanisms in the nucleus—affecting chromatin state, transcription factor availability, and noncoding RNA crosstalk—LNCAROD is considered a potential therapeutic target, but presents safety and efficacy challenges because of its fundamental role in gene regulation and nuclear compartmentalization[2][3][4].
