Long non-coding RNA ITSN1-2 (lncRNA ITSN1-2) is a regulatory RNA molecule that plays a significant role in the pathogenesis of inflammatory and ischemic diseases (Nie and Zhao, 2020) [7.2.4]. It primarily functions as a competing endogenous RNA (ceRNA), acting as a 'sponge' for microRNAs such as miR-125a, miR-195-5p, and miR-2683-3p (Yue et al., 2019; Bian et al., 2025) [7.3.2, 7.4.3]. By sequestering these miRNAs, lncRNA ITSN1-2 prevents the silencing of target mRNAs like IL-23R and NOD2, which drives pro-inflammatory signaling pathways including NF-κB and NOD2/RIP2 (Nie and Zhao, 2020; Yue et al., 2019) [7.2.4, 7.3.2]. Its upregulation is strongly correlated with disease severity and poor prognosis in conditions such as Inflammatory Bowel Disease (IBD), Rheumatoid Arthritis (RA), and sepsis (Zeng et al., 2019; Li et al., 2019) [7.2.3, 8.1.2]. Due to its involvement in these pathways, lncRNA ITSN1-2 is considered a promising therapeutic target and a diagnostic biomarker for monitoring disease activity (Huang et al., 2021; Bian et al., 2025) [8.2.2, 7.4.3]. It is also being explored in the context of ischemic stroke and severe acute pancreatitis, where its levels serve as indicators of tissue damage and inflammatory response (Ren et al., 2020; Li et al., 2019) [7.1.4, 8.1.2].
