Long non-coding RNA NONRATT021972 is a regulatory RNA molecule identified in rat models that plays a significant role in the pathogenesis of diabetic neuropathic pain (DNP) (Liu et al., 2016, Molecular Medicine Reports). It is notably upregulated in the dorsal root ganglia (DRG) of type 2 diabetic rats, where its increased expression correlates with the development of thermal hyperalgesia and mechanical allodynia (Peng et al., 2017, Purinergic Signalling). The molecule functions by positively modulating the expression and mediated activation of the P2X3 receptor, a key purinergic receptor involved in transmitting nociceptive signals (Yu et al., 2020, Neuroscience Letters). Research has demonstrated that silencing NONRATT021972 using small interfering RNA (siRNA) can effectively decrease P2X3 protein and mRNA levels, leading to a reduction in pain-related behaviors in diabetic models (Liu et al., 2016, Molecular Medicine Reports). While there are currently no approved clinical drugs targeting this specific lncRNA, it is considered a promising therapeutic target for the treatment of chronic neuropathic pain associated with metabolic disorders. Its role as a regulatory upstream element of the P2X3 pathway offers a novel approach to pain management that may bypass some of the limitations of direct receptor antagonism.
Experimental inhibition via RNA interference (siRNA) or antisense oligonucleotides to downregulate lncRNA levels, which subsequently reduces the expression and activation of P2X3 receptors in dorsal root ganglia (Liu et al., 2016, Molecular Medicine Reports).
