Overview

Long non-coding RNA osteoblastogenesis associated 1 (lnc-OB1) is an osteoblast-enriched lncRNA that is upregulated during osteoblast differentiation[2][5]. Its overexpression enhances osteoblast activity, bone mass, and mineralization in mouse and human models, whereas its suppression reduces these processes. Mechanistically, lnc-OB1 promotes the expression of the osteogenic transcription factor Osterix (OSX), likely by epigenetically reducing H3K27me3 marks on the OSX promoter[2][5]. lnc-OB1 is downregulated in osteoblasts from older osteoporotic humans and mouse models of osteoporosis, but its targeted overexpression confers resistance to bone loss, making it a promising therapeutic target for osteoporosis[2][5][1]. There are, as of now, no approved drugs that act directly on lnc-OB1, but experimental therapies delivering this lncRNA to osteoblasts are under investigation in preclinical models.
