Overview

Long noncoding RNA NONRATT021972 is a regulatory RNA molecule that plays a critical role in the development of diabetes-related complications, particularly diabetic neuropathic pain (DNP). It is significantly upregulated in the dorsal root ganglia (DRG) and blood of type 2 diabetic subjects, where its expression levels correlate positively with the severity of neuropathic symptoms. Mechanistically, NONRATT021972 promotes inflammation by increasing the production of tumor necrosis factor-alpha (TNF-alpha) and modulates the expression of purinergic receptors, such as P2X7 and P2X3, which are involved in pain transmission. Beyond neuropathy, it has been implicated in diabetic cardiac autonomic neuropathy, myocardial ischemia, and hepatic glucose metabolism through the regulation of glucokinase. Experimental studies using siRNA-mediated knockdown have shown that reducing NONRATT021972 levels can attenuate inflammatory responses and alleviate pain behaviors in diabetic models. Consequently, this lncRNA is considered a promising therapeutic target and a potential diagnostic biomarker for managing chronic complications of diabetes.

Mechanism of action

The primary mechanism involves the use of small interfering RNA (siRNA) to silence the expression of NONRATT021972. This knockdown leads to a reduction in the pro-inflammatory cytokine TNF-alpha and the downregulation of P2X7 and P2X3 receptors in sensory neurons, which effectively inhibits the transmission of nociceptive signals and reduces neuropathic pain.
