Overview

Loricrin cornified envelope precursor protein is the primary structural protein of the cornified envelope in the epidermis, comprising over 70% of its composition. It is critical for the formation and mechanical strength of the stratum corneum through protein cross-linking, enabling skin barrier function against environmental insults. Loricrin is a highly insoluble protein rich in glycine, serine, and cysteine residues, expressed exclusively in terminally differentiated keratinocytes in the upper layers of stratified epithelial tissues such as skin, oral and anal mucosa, esophagus, and vagina. Mutations lead to skin disorders like Vohwinkel syndrome and palmo-plantar keratoderma. Its altered expression is associated with dermatological diseases and has utility as a biomarker in the assessment of keratinization and potential malignant transformation in some mucosal lesions
