Lotrafilcon A is a high-performance silicone hydrogel material primarily used in the manufacturing of soft contact lenses, such as the Focus Night & Day and Air Optix brands (FDA PMA P980006). It is a biphasic copolymer consisting of a fluoro-silicone phase that provides exceptionally high oxygen permeability (Dk of 140) and a hydrogel phase that facilitates the transport of water and ions (Contact Lens Spectrum, 2008). This unique structure allows the material to maintain corneal health by ensuring adequate oxygenation, even during extended or continuous wear for up to 30 nights. Beyond vision correction for refractive errors like myopia and hyperopia, Lotrafilcon A is frequently employed as a therapeutic bandage contact lens to promote epithelial healing and provide pain relief in conditions such as corneal erosions, bullous keratopathy, and post-surgical recovery (NIH, 2003; NIH, 2005). As a synthetic polymer, it is not a biological target like a receptor or enzyme but rather a medical device component that interacts physically with the ocular surface and tear film. While it does not have a biochemical mechanism of action, its physical properties are critical for its clinical efficacy in both refractive and therapeutic applications.
